A couple of techniques I’ve found go well with the SC protocol:

 

1. Occlusion bands. These are said to increase testosterone and growth hormone. I was skeptical at first, but soon became a believer. Two minutes is better than a visit to the gym. I use this separately from the protocol, but they appear to be synergistic with it, even just using them on my arms.

 

2. Skin brushing with niacin fission. This involves —

 a) One gram of niacin, which produces fission and massive flushing,

 b) A hot shower,

 c) Skin brushing with a boar bristle brush. I use Goody Straight Talk Boar Styler Hair Brush. It’s very stiff. I bought a couple at Walmart for $8 each. And optionally,

 d) Senolytics like curcumin and quercetin.

 

Combining niacin, skin brushing and a hot shower is amazing at rejuvenating and tightening the skin. Far better than derma rolling.

Note: Don't use this much niacin unless you are already familiar with the effects.

Sounds a little intense but I will give it a go at some point. What's the thoery, is the degree of the flush key here? I found flushing to be a capricious fellow, the incubation period and intensity would be quite varied. I did remember reading why the flush can be so inconsistent but don't recall the reasons, so perhaps there are pre-treatment protocols that might intensify the effect?

 

I oddly enjoyed the experience, often feeling somewhat mentally refreshed afterwards, if a little cold.

The niacin flush is subject to tolerance, but the tolerance is quickly lost. If you allow a day or two between treatments, that may be enough. The more severe the flush, the better it seems to work. And I don't find it in any way painful. It's the pleasurable feeling of scratching an itch, but highly magnified.

There are ways to reduce the flush, which might in turn provide the answer. It might also offer an explanation as to why my effects were so varied: histamine. I have certainly had problems with histamine which I put down to zinc deficiency. I ran in to real problems a few years back taking high doses of NR and N+R with histamine (from the N) and uric acid with the R. I couldn't touch any Nicotinamide or Nicotinamide Riboside without a bad reaction but was fine with nicotinic acid/niacin.

nb I found this recent paper while trying to see if niacinamide increases increaes PGD2, doesn't seem relevant but might be of experimental interest. I may research a little more thoroughly another time.

Body and brain mito fusion brownies

For use with the Stem Cell protocol

 

Betty Crocker Fudge brownie mix* (519 g)

2 eggs

4-5 tbsp water

30 g (4 tbsp) Sunflower lecithin powder  

30 g (7 tbsp) Dihydromyricetin powder

120 g stearic acid

 

*Or any mix that requires ½ cup oil, but don’t use the oil. Diet versions don’t work well.

 

Mix with power mixer and bake as directed on the box. Cut into 16 pieces, dust with flour to avoid sticking, and freeze. Dosage: 1 piece three hours before SC treatment. The dihydromyricetin degrades the taste a bit, but freezing improves it. The lecithin is to make the dihydromyricetin more bioavailable and to supply phospholipids for new brain cell myelination.

 

I’ve noted before that fusion brownies can raise BP, but adding lecithin appears to eliminate that.

 

Hi Turnbuckle. Thanks for sharing this. I guess "SC treatment" refers to #1619. Can the ingredients in #1619 be cooked in brownies as well? (or maybe pancakes?) I don't know whether it's Dihydromyricetin itself or my source of it, I found mixing it into beverage makes it undrinkable.

Thanks James. I did come across this product on Amazon. But each capsule is 300mg. For this new brownie addition Turnbuckle posted, it would require 100 capsules. Even for the SC protocol, the minimum dosage of 2g is 6~7 capsules. And the price value is significantly lower than powder products.

 

Thus so far I've been trying out powder products mainly:

They are of different colors. The first one is white while the second is brown. So I was wondering what's the better variation.

Only if you still have follicles with viable stem cells. When I first started using C60 I did grow more hair that filled in my MPB, but then that reversed, and apparently permanently. Using C60 without fusion will ultimately deplete stem cell niches, and the protocol needs something to work with.

Correct me if I'm wrong. It seems to me that this stem cell with senolytics protocol ought to repair damage to slowly dividing cell tissues and organs such as the heart. For example, myocarditis.

OK, well I had a go at the niacin/hot shower/strong brush combo. Quite brutal. And I only used a strong nail brush! It was just a few hours ago, I'm not sure I would say I look different but my skin is smoother and less dry, noticeably oilier I would say. I never brush my skin so whether it could be that alone, idk. I have derma-rolled before, as have you I believe - with lysine solution some years ago (?) - have you not thought to subsitute into a derma-roller for the brush? After you, obviously, though.

 

It does seem as per the link below that excess PGD2 on the skin surface causes hair loss and whether or not that might be stage 1 of trying to regain hair. It seems from the articles linked that PGD2 isn't released from the mast cells since there is no uptick in histamine during a flush but I assume the PGD2 mediating the flush was 'pre-released'. 

 

I know though that hair regrowth can be achieved, just not where I need it the most. I have lost and regained hair several times over the last 8 years since taking c60 (though I believe i noticed through other supplementation NR, zinc) on my legs. But also temple hair, which I believe followed from a large dose of senolytics or possibly one of your therapies - it was reported by someone else too independtly. I have noticed some reversal of grey in bounded-growth-type hair a few years back and in the unbounded stumble, though I thought I may have seen more stubble for a while on the back of c60.

 

Anyhow, my anectdotal experience seems to demonstrate that there is something recoverable about hair that knows its limits. Also the incident of the recovery temple hair given its proxmity to the follicules surrendered long ago was interesting, I thought, and that it was a repeated observation.

So speculating, I wonder if it is possible that the could be a period where in fact stem cells are still there but inhibited in some way. Perhaps, there are different thresholds with different hair type.

I recommended the brush treatment as it not only worked, but felt great. I wouldn't recommend it to anyone who found it brutal.

 

As for use of C60 without fusion, I expect that this will deplete stem cells, and when they are gone from a hair follicle, expect no return. Though as you say, there may be some other reason they've gone quiescent.

Niacin provides mito fission, which is necessary for apoptosis and for SC differentiation, and the combination of niacin and heat dilates the blood vessels to a maximum, making the skin stiffer and thus more easily abraded by a suitable brush. The appropriate brush will depend on the thickness of your skin. A more aggressive brush can be used on thicker skin.

Thanks for the explanation. Unfortunately this experiment was rather confounded with subsequent fresh NMN dosing yesterday; however, the oiliness of the skin seemed to occur quite quickly after the protocol (which was without seonolytics). During the night the texture felt the same, I rarely put stuff on my skin but occasionally have done so with c60 before bed recently and I had to question myself half asleep as to whether I had applied c60 as the feeling was quite similar.

 

Today I found myself itching quite intensely around the shin and calf, where predominatly the recurring hair loss occurs - this has happened before post nicotinic acid, but I never paid attention to it. So now I'm beginning to wonder if this is about PGD2 being liberated and whether or not this has contributed to dry skin (as reported I have had some significant intermittent problems with histamine over the last couple of years, the residual effects of which might there in the surface of my skin). I have decided I will give the Sorbic Acid solution ago at some point, maybe if I'm lucky I'll slow down some of the hair loss.

 

Anyhow, I appreciate this has diverged. I will post an update but perhaps create a fresh thread if reults are interesting. 

 

edit: might this may explain the post niacin flush itching though eliminating cutaenous PGD2?

Quick aside since we are discussing stem cell renewal...

 

Not a huge fan of long fasts but this impressed me enough to rethink that stance:

 

Fasting triggers stem cell regeneration of damaged, old immune system (Studying chemo patients):

 

In the first evidence of a natural intervention triggering stem cell-based regeneration of an organ or system, a study in the June 5 [2014] issue of the Cell Stem Cell shows that cycles of prolonged fasting not only protect against immune system damage — a major side effect of chemotherapy — but also induce immune system regeneration, shifting stem cells from a dormant state to a state of self-renewal.

 

The study has major implications for healthier aging, in which immune system decline contributes to increased susceptibility to disease as people age. By outlining how prolonged fasting cycles — periods of no food for two to four days at a time over the course of six months — kill older and damaged immune cells and generate new ones, the research also has implications for chemotherapy tolerance and for those with a wide range of immune system deficiencies, including autoimmunity disorders.

 

They noticed that "marginal" stem cells were getting cannibalized during the fast and then there was a refresh of new, fresh stem cells to pick the WC back up. This seems to track with fasting activating ISR and other conservation steps.

 

Safety note: they call out that during the fast WC dropped - stop the fast if you get sick.

 

So the big question of course, @Turnbuckle, would this synergize or compete?

I have been following the protocol once every 14 days for 5 months (a total of 10 cycles). My data is expected to be ready March 1st (although the 1st test was ready about 1/2 that time). Upon receipt of the data, I will post the exact details of what I took and dosages (although it almost exactly mirrors Turnbuckle's)

Before starting this protocol, I was taking C60 every day.  I switched to taking it once every 14 days for the protocol, but I did not like how I felt, so I have been taking it once per week for the last 3 months.

1. My dad and both my grandfather's did not have full gray hair until they were ~65.  I have a few grays (maybe 30 total) but not many, so I do not think they are a good marker.  Skin has been noticeably softer and does not crack at all any more.
2. I have been really taking care of myself for the past 20ish years.  I can confidently say, today I feel better than I did 25 years ago.  As far as feeling better after starting the protocol, not huge.  I have been taking C60 for many many years now, and I can say after taking C60 for the first time all those years ago, I felt something way more significant then. 
3. So I track my sleep via fitness watch religiously.  My average sleep before starting just C60 was 6 hours and 35 minutes the 5 months before I started taking it.  5 months after starting just C60 was 8 hours and 21 minutes.  5 months before starting the Turnbuckle Protocol my sleep was average 8 hours and 23 minutes.  The 5 months of doing this protocol my sleep has been 8 hours and 34 minutes.  Average of the past 3 years 2019, 2020, 2021; 8 hours 24 minutes.
4. My blood pressure stayed the same ~120/80.
5.  Yes, I also do not seem to get a dehydrated feeling as much.  I do track my water intake, and I average 60-80 ounces a day.
6. I continue to Run or Hike 3 miles a day 5 days a week (sometimes up to 20 miles on the weekend).  Really see no difference.
7. When I started the protocol, Part 2 "Senescent Cell Replacement"  I felt drained and unable to function for ~24 hours the first time.  Then it reduced by ~4 hours each time.  The last 4 or 5 times it did not drain me at all (I am hoping that is because I drained the majority of the Senscent cells...).   I have only ever done Part 2 once per 14 day cycle.

Diet:
-- What I deem "Healthy Keto."  Majority Veggies with 3-6oz of Meat per meal.  Mix in healthy fats, avacado, coconut oil, nuts, and olive oil.

This protocol is designed to proliferate SCs and thus prevent depletion. Nevertheless, SC telomeres may shorten with replication and ultimately lead to cell cycle arrest. SCs produce telomerase, but this may not be enough to keep them from shortening with repeated replication.  You can attempt to reverse telomere erosion, but you have to be judicious about it. I suggest 1 cap (500 mg or so) of astragalus extract with the protocol on an infrequent basis. Maybe once every fourth treatment or so. If you do too much, the replicative life of somatic cells and the intermediate transit-amplifying cells (TACs) will be extended and their replacement by SCs blocked. Then you will see your epigenetic age go up instead of down. One cap may not seem like much, but given that SCs already produce telomerase, you need to boost it only a little.

That's what I thought.

 

I was wondering if there is a way to selectively increase SC telomeres but not those of regular cells. But if what you say is the case, that SC's make more telomerase than regular cells, then the selectivity is already built in. Just use a sufficiently small amount of the astragalus extract such that only the SC's benefit from it and that's the selectivity.

Welp, I recieved my methylation age test back from https://www.mydnage.com . All I can say is that I am appalled... It came back as +9 years over my actual age. I don't even know what to do with this; going from +4 to +9 is devastating. Needless to say, I will discontinue the regiment.

Here is the regiment.  I did it for 5 months without fail.  One cycle every 14 days
 

Part 1: Stem cell self-renewal
Time 0 — 6:00 am day 1

  • Kokum Butter (50-60% Stearic Acid) — ~10 g (food grade, in brownie with lecithin)

  • Ox Bile — 500mg

  • Betaine Hydrochloride — 2 g

  • Digestive Enzymes — 2 g (particularly Lipase)

Time 3:00 — 9:00 am day 1

  • Sulforaphane — 100 mg

  • Brown Mustard Seed — 500 mg

  • Liposomal glutathione — 1 g

  • SAM-e — 500 mg

  • Resveratrol, 50% Trans — 1g

  • AAKG  — 2g

 
Time 3:30 — 9:30 am day 1

  • C60 — 3 mg (in oil) aka 1 tsp

  • Threonine — 2g

  • Lysine — 2 g

  • Methionine — 1 g

  • Leucine — 1 g

 Time 5:30 — 11:30 am day 1

  • Lysine — 2 g

  • Methionine — 1 g

Time 7:30 — 1:30 pm day 1 (can repeat at time 9:30 if needed)

  • Lysine — 2 g

  • Methionine — 1 g

Part 2: Senescent cell replacement (24 hours later or more)

Time 30:00 — 12:00 pm Noon day 2

  • Lysine — 2 g

  • Methionine — 1 g

  • Heaping tablespoon of Coconut oil

  • Nicotinamide — 1.5 g

  • D-Ribose — 1.5 g

  • NMN — 1 g

  • Curcumin  — 2g

  • Bioperine — 30mg

  • Quercetin Phytosome with Bromelain —500mg

  • (maybe add Fisetin in the future)

Time 32:00 — 2:00 pm day 2

  • Lysine — 2 g

  • Methionine — 1 g

Time 34:00 — 4:00 pm day 2 (can repeat at time 36:00 if needed)

  • Lysine — 2 g

  • Methionine — 1 g

I am a chemist from Russia, Moscow. Your methylation results are frustrating. I greet you and want to help you.
 
It seems to me that you are faced with a systemic problem that cannot be solved by the stem cell renewal protocol alone.
 
Methylation processes seem to be influenced by proteins and the extracellular matrix. I commented on this post:
 
 
Aging proteins can be partially removed by plasmapheresis.
 
Perhaps the elasticity of the extracellular matrix can be restored with the help of ground dill seeds and hyaluronic acid.

My set of supplements:
 
1. Ubiquinol - 200 mg / day - to increase the production of active substances.
 
2. Boiled ground Dill seed fruits brewed with boiling water - 3-4 g / day - 300 mg of boiling water in a thermo mug and infused for 1 hour (minimum). After that, the warm broth is poured into a regular mug, without overflowing the sediment (decantation). The decoction allows the production of elastin components. Tested on mice and tested by Russian biohackers (including me).
 
3. Hyaluronic acid - 100 mg / day + 600 ml of pure water. Must be taken 2 hours after a meal (minimum). Or 40 minutes before a meal. I generally drink before bed.
 
4. Nattokinase (2000 FU) - 2 to 4 capsules per day. Thins the blood. The goal is to facilitate the delivery of elastin and hyaluronic acid components to the extracellular matrix and their incorporation into the matrix.
 
5. Inositol hexaphosphate (IP 6) 2-4 capsules per day (I did not take these capsules every day) - a substance that forms complexes with calcium, thus removing excess calcium from the extracellular matrix. The goal is to reduce the rigidity of the extracellular matrix and remove excess calcium from there, which should not be there.
 
6. Trainer (ellipse) for 40 minutes a day - 5-6 days a week - at a pulse rate equal to 65-70% of the maximum allowable for your age. The goal is to improve blood microcirculation, increase the elasticity of the extracellular matrix, physically adjust the blood to tissue areas that are difficult to supply with blood.
 
Two key ingredients are ground dill seeds and hyaluronic acid. Without them, the protocol will not work. 
 
Without changing the elasticity of the matrix and plasmapheresis, you are unlikely to achieve systemic improvements. I assume that after system improvements you will be able to continue your protocol with better results.
