Regarding the studies mentioned at the links below, it seems Alpah-ketoglutarate might be involved in lifespan extension by inhibiting ATP synthase and TOR.

https://www.ncbi.nlm...pubmed/24828042

https://medicalxpres...n-nematode.html

I have been taking 250 MG of Niagen per day and I was thinking of taking arginine alpha-ketoglutarate the "Real AKG" brand, in a supplement from swansons shown at the link below.

https://www.swansonv...utarate-90-tabs

Real-AKG brand

http://www.innlabs.com/AKG.html

When I saw the study on possible lifespan extension via AKG, and the likely cause, it got me wondering if AAKG is compatible with Niagen and an increase in NAD+ ?

Would AAGK likely oppose the effects of Niagen or help it  somehow ?

I would appreciate any thoughts.

Thanks,

John

P.S. Sorry about the spelling of Nicotinamide Riboside in the post title, I copied & pasted it from somewhere and realized it was spelled wrong after the post. Perhaps a MOD can correct it.

I think it is very risky to block ATP production with Alpah-ketoglutarate. Cyanide poison works by blocking ATP production.

NR needs ATP to convert to NAD+. So it is incompatible with NR.  NR is a much better fasting mimic than everything else.

mTORC1 is not an ideal anti aging target because it is modulated differently in different cells. Side effects include reduction in testosterone, damage of testis, suppression of intestine immune cells and

suppression of intestine stem cells.

Can anyone explain the following to me in simple language? What does the word "rescue" mean in the following context?:

"Alpha-ketoglutarate rescues ATP production, GSH levels, and cell survival during glutamine withdrawal in cells lacking the Rb family. TKO cells were cultured in glutamine-free medium for the indicated time periods with or without addition of 7 mM dimethyl-α-ketoglutarate. (a) Viable cell number was determined by hemocytometer enumeration after trypan blue staining. Data are represented as mean viable cell number±s.d. from triplicate measurements from three independent experiments. *P<0.005. (b) Steady-state ATP was determined between WT and TKO cells as described in the Materials and methods. Data are represented as μM/mg protein, and shown are mean±s.d. of triplicate readings from two different experiments. *P<0.01. © Representative flow cytometry histograms of TKO MEFs cultured in complete medium, medium lacking glutamine, or medium lacking glutamine with the addition of α-ketoglutarate for twenty-four hours. (d) Mean fluorescence intensity in TKO MEFs cultured in the absence or presence of glutamine or α-ketoglutarate. Data are presented as the mean±s.d. from triplicate readings from two separate experiments. *P<0.01. (e) GSH levels in TKO MEFs after culture in indicated medium conditions for 24 h. Data are represented as relative GSH levels between the different conditions with+GLN sample set to 1, and shown are the mean±s.d. from triplicate measurements from two separate experiments."

You are making very broad assumptions without evidence to back them up. As ususal.

Metformin for example, is also a mild toxin to the mitochondria and works by reducing ATP production. Mitohormesis from mild "poisoning" by the botanically derived metformin could explain some of the beneficial effects of that molecule.

This is similar to what I was thinking. So in your view is it ok to take nicotimomide riboside (and also straight nicotimomide) alongside taking AAKG?

I wouldn´t take either. NR because of no evidence that it really does anything in healthy people and AAKG because of lack of both efficacy and longterm safety as a supplement.

I don’t know about NR but plain nicotinamide/niacinamide can prevent skin cancer:

https://www.ncbi.nlm...les/PMC4570055/

And the efficacy and safety of alpha-ketoglutarate seems supportable on both counts. I can’t find any research that says it has no health benefits or that it’s potentially dangerous.
