How long to stay on/off Rapamycin to not inhibit mTOR2?

The benefit of preventative Rapamycin therapy seems to be inhibition of mTOR1, but side effects and negative effects can come when one simultaneously inhibits mTOR2 by staying on Rapamycin too long.  Does anyone have any research for how long to stay on Rapamycin before mTOR2 becomes downregulated?  Also, how long should one stay cycled off Rapamycin before getting back on it so that one could only downregulate mTOR1 and not mTOR2?

Hi.  I'm pretty new at all this.  I started Sirolimus a month ago.  My god, what a difference!

 

My 4 siblings all have ulcerative colitis.  I have something less but still related issues.  They don't concern about diet, exercise, etc.  I've been a health nut forever.  Now following Michael Greger's advice very closely.

 

So now with Sirolimus, the condition disappeared!  There is absolutely no sign of UC.  It is as if I've never had it at all.  My only prior colonoscopy revealed aphtha (chancres) the gastroenterologist said was too mild to be UC.  It didn't feel that good.  No question they are not there anymore.

 

In the spirit of this thread, I'm wondering about this regimen.  I'm taking 6mg 1x/wk.  Is there some longterm risk?
