If this was so important how come he never told us sooner?

 

Edited to add the following:

 

I just found a video where he says he takes resveratrol daily but every couple of weeks has a day off from taking it to give his liver a rest. (Why would he want to give his liver a rest? Is resveratrol bad for the liver?)

 

So in the video above he says resveratrol should be taken on alternate days, and in the video below he says he takes it daily... I'm confused.

This appears to be the study Sinclair is referring to in the first video above. The study says:

 
"Several studies have reported that resveratrol does not extend the lifespan of mice when provided in a standard chow ad libitum. However, resveratrol does extend mouse lifespan when provided in a high calorie diet or in standard chow fed to mice every other day."
 
 
Yet in another study resveratrol was given daily:
 
"To each of the diets, we added resveratrol at two concentrations that provided an average of 5.2 ± 0.1 and 22.4 ± 0.4 mg kg−1 day−1, which are feasible daily doses for humans."
 
 
Translation:
 
Each of the diets they tested, they added resveratrol at two different amounts. These amounts provided an average of 5.2 ± 0.1 and 22.4 ± 0.4 milligrams per kilogram of body weight per day. These doses are considered feasible and safe for humans to take on a daily basis.
 
So again, why is Sinclair taking resveratrol daily? What's going on?
 
Or maybe the "every other day" in the first quote above is referring to the frequency of the the chow and not resveratrol. On reflection, this seems likely. 
 
This brings up another question, though: are resveratrol's life extension benefits based on the diet you are on when taking resveratrol?
 
The first quote above says the effects of resveratrol on extending lifespan in mice depended on the diet the mice were fed. When mice were provided with resveratrol in their regular diet without any restrictions, it didn't extend their lifespan. However, when resveratrol was given to mice in a high-calorie diet or when they were fed regular chow every other day, it did extend their lifespan.
 
It appears that the effects of resveratrol on lifespan extension in mice are influenced by both diet and caloric restriction (the latter, bizarrely, seeing as resveratrol is meant to mimic CR). The studies show that when mice are given resveratrol in their regular diet without any restrictions, it does not extend their lifespan. However, when resveratrol is administered to mice in a high-calorie diet or when they are subjected to alternate-day fasting (where they are fed regular chow every other day), it does extend their lifespan.
 
So the benefits of resveratrol in extending lifespan may require specific dietary conditions or caloric restriction to be effective.  Maybe these studies are specific to mice and may not directly translate to humans.

I found the reason why Sinclair takes a day off every couple of weeks from taking resveratrol daily (see the study below). He takes 1 gram, though, which is quite a lot, so maybe he needs to take a break. Maybe people who take less don't need a break.

We investigated the dosing-time dependency of acute resveratrol administration on lipoperoxidation level found in the heart, liver and kidney of male rats synchronized with a 12-h dark-light cycle. Resveratrol was administered by the i.p. route at the middle of the dark (6 h after dark onset, HADO) or light span (18 HADO) and thiobarbituric acid reactive species (TBARS) measured 4 h later at 10 and 22 HADO, respectively. Basal TBARS levels in the three organs were higher during the night span when compared to day span. Resveratrol effect on tissues TBARS was also dosing-time dependent. When administered during the dark phase, resveratrol decreased TBARS levels whereas at the light span, the polyphenol increased TBARS in the three organs. Resveratrol behaved as an antioxidant during the dark span and as a pro-oxidant during the light span. I watched a interview on Youtube an David Sinclair was forced to answer this question . Do you take Rampymcin an he said On occasion . His facial expressions told me he wasn't happy answering the question .

I take it once a week 500mg, once a week is much more tolerable. Biggest problem of the substance is that it lowers too much estrogens, and also at low concentrations, so that makes joints hurt and other trouble. But using it just once a week no big problem.
