Three doctoral candidates from Stanford University have jointly exposed the "stem cell black market" in the United States to the public media. Chinese scientists have discovered that the activation of endogenous viral elements in the human genome is a key factor in inducing cellular and organ aging. They cultured and screened millions of human cells, validating dozens of previously undiscovered genes that promote cellular aging from over 20,000 human genes. Through repeated screening, they ultimately identified a novel human pro-aging gene named KAT7. Experiments confirmed that partially inactivating this gene in the livers of elderly mice allowed 81% of the mice to live beyond 130 weeks, which is approximately equivalent to 80 years in human age.

The journal Nature simultaneously published two papers from Xiamen University. They found that a metabolite called lithocholic acid (LCA) plays a key role in the anti-aging effects of "caloric restriction." This discovery suggests that by supplementing with lithocholic acid, one may achieve the "anti-aging" benefits of caloric restriction without having to give up delicious food or subject oneself to hunger.

Exogenous LCA has not been proven to be safe for humans and further research is needed. The main value of these papers is the understanding of the AMPK pathway.Its value is not to extend lifespan, and the AMPK pathway itself is not a pathway for extending lifespan

According to the independent panel of experts, while these essential nutrients are important for overall health, there is not enough evidence to suggest a net benefit for these endpoints, and supplementation may be associated with the risk of developing kidney stones.

The average life expectancy for Chinese men in 2024 is 69.9 years, and for Chinese women, it is 78.5 years.The average life expectancy of Chinese men in 2023 is 73.64 years old. 69.9 is a very small data; I know the average life expectancy of Americans is around 80 years old."

In 2024, the Center for Drug Evaluation and Research (CDER) of the US Food and Drug Administration (FDA) approved a total of 50 new drugs, including 34 new molecular entities and 16 biologics. Among the new molecular entities, small molecules predominated, accounting for about 91% (31), with the remainder consisting of nucleic acid and peptide drugs; among the biologics, 81% were antibody-based (10 monoclonal antibodies and 3 bispecific antibodies), with the rest being fusion proteins and Type A botulinum toxin. It appears that small molecules are still the main force in anti-aging efforts.

President Trump wants to annex Canada, the Gulf of Mexico, Greenland, and the Panama Canal to the United States, and I fully support this move. In fact, if it were possible to include China and other countries around the world into the United States, forming a single nation on Earth, that would be even better. This would eliminate many barriers for the anti-aging geek community, including language barriers, currency obstacles, cultural habits, ways of thinking, and educational levels. On January 6th, the United States reported its first death from the avian influenza virus (H5N1). Although no human-to-human transmission has been detected, it is crucial to be vigilant about potential viral mutations. We are currently in an era prone to viral outbreaks for two main reasons: one is natural factors, as viruses periodically emerge that can influence human DNA and drive evolution; the other is human factors, with the novel coronavirus being a coronavirus engineered from HIV. In the future, viruses may become the third leading cause of death for humans. The United States has banned TikTok, and the confrontation between the U.S. and China is intensifying. Despite issues with China's system and bureaucracy, it is the ordinary Chinese people who are affected by this confrontation, while some high-ranking Chinese individuals actually hold U.S. citizenship(most CBC is CBA). We hope that the confrontation between China and the U.S. does not affect humanity's pursuit of combating aging.

A study published in Cell Metabolism magazine indicates that exosomes rich in miR-302b, known as hESC-Exos, can reverse the proliferative stagnation of senescent cells, promote cellular rejuvenation, and restore vitality to aged mice. The research found that miR-302b, by targeting Cdkn1a and Ccng2, restores the expression of cell cycle-related genes, increases cell division, and improves the physiological functions and appearance of aged mice. Long-term delivery of miR-302b can extend the lifespan of mice, ameliorate signs of aging, without increasing the burden of tumors or diseases, effectively slowing down the aging process.

A study published in a Nature journal has indicated that the metabolic product of gut microbes — phenylacetylglutamine (PAGln) — increases with the age of the host and accelerates the aging process of the host. The research found that PAGln activates the adrenergic receptor-AMPK signaling pathway, increases the level of reactive oxygen species (ROS), mediates DNA damage, and leads to cellular senescence. The researchers proposed that the use of adrenergic receptor blockers (carvedilol) and Bcl-2 inhibitors (ABT263) can counteract PAGln-induced cellular aging.

Recently, an article titled "Somatic mutation as an explanation for epigenetic aging" has become popular. The article suggests that the main approach of Altos Labs, which focuses on epigenetic reprogramming, might be incorrect. This aligns with the main anti-aging principle of LongevityDiary since its inception, which is that the primary cause of aging is errors in DNA. We have never been optimistic about the direction of in vivo reprogramming using osk/oskm. Even though Altos Labs has invested $3 billion, we believe that the approach is incorrect. LongevityDiary‘s method of correcting DNA errors is correct and has not been overturned to this day.

The overall evidence suggests that CR, or low-carb diets and other methods of weight control, may not be a good anti-aging strategy. This conclusion is based on the following facts:

 

  1. Chinese star Barbie Hsu, as a celebrity, had strict weight management and died at the age of 48.
  2. The general practical experience of ICU doctors is that overweight patients who enter the ICU often survive and become thinner, while thin patients who enter the ICU have a high probability of death.
  3. Liu Jiaxin, a famous low-carb advocate in China, nicknamed "Old Hippo," had a 28-year history of diabetes and experienced multiple severe complications, including arteriosclerosis, two heart attacks with stents, diabetic foot, diabetic neuropathy, and diabetic retinopathy. At that time, Jiaxin's condition was very critical, and insulin injections of 100 units were ineffective. Even the Peking Union Medical College Hospital refused to admit him. Later, he reversed his condition through a ketogenic low-carb diet, no longer needing any medication, losing 50 pounds, and experiencing a significant reduction in carotid artery plaque. He recently passed away due to a sudden illness at the age of 62.
  4. In Jan Gruber's new paper, they evaluated mortality data to create weights for the new mortality calculator LinAge2, which found that BMI has a protective effect on the mortality of both men and women. The higher the BMI, the longer the lifespan.
  5. The famous American diet (a diet high in fat) has led to Chinese people generally being slimmer than Americans. The average life expectancy for American men is 75 years, which is 5 years longer than the average life expectancy for Chinese men, which is 70 years.
  6. The elderly are prone to sarcopenia and are often concerned with how to increase their weight rather than lose weight.

Perhaps the benefits of weight management techniques such as CR (Calorie Restriction) are only temporary, and in the long term, they may be harmful.
