What is the half-life of this stuff, and are there active metabolites? 3alarm, it could have been downstream effects or a buildup effect instead of a bunch of synergistic compounds together. I don't know too much about this and I'm just throwing that out there, for what its worth. I bought a little of this RLP-47-53-1 and I've got memory problems of my own, but want to wait a while before I try it out of caution.


Interesting thought actually. I'm not sure exactly. Hansl's paper did indeed mention active metabolites, I believe one of them was hydroxylated.


NOTE ADDED IN PROOF
Following completion of this study it was found that the particular lot of PRL-8-53 that had been used contained a small amount of the desmethyl compound as admixture, which may also have contributed to the observed effects. However, since this compound is considered a metabolite of the parent compound (8-53), its presence should not detract from the findings

I ordered some Coluracetam (1g) and I will try out if there is main effects of Coluracetam alone on the scores on my WM or if Coluracetam and PRL-8-53 together have synergistic positive effect on the results across test presented on Cambridge Brain Sciences homepage.

Side note: in my case PRL-8-53 seem to have very strong effect on LT memory. While I am studding for exams I usually read through the slides and lecture notes and highlight the most important facts and concepts. The seccond and third time I go through notes and slides I only concentrate on highlighted parts of my materials. So I have pretty good intuitive feel of my usual level of memorization after initial reading of materials - while on PRL-8-53 I seem to remember much much more after reading the materials first time.

So as I mentioned in my previous post, I was taking 5-10 mg of PRL every day over the past week and a half or so. I did not take any PRL Saturday and this Sunday I have had a headache all day which is pretty unusual for me. Maybe taking it every day is not the best way to go about using PRL. Other people seem to be doing quite well with it, but I thought I would mention this in case other people find the same result.

I've been taking PRL-8-53 twice a day since Sunday. I presume I am taking 10mg with each dose as 5 measues from the supplied spoon from NSN weighs about 50mg on my scales. I am not taking any other nootropics at the moment.

I'm quite liking the results so far. I seem to be able to concentrate and focus a little better at work (I develop software). It's not a stimulant for me - and I'm not looking for a stimulant anyway. Green tea/ginseng/kola nut/yerba mate are all good enough for me. I have found some sleeping problems though, but these diminished a little if I don't have an afternoon tea. i'll keep experimenting with it over the next couple of weeks and then I'll try the IRDA-21 that I also have.

As a side note: I'm in Canada and Canada customs opened the package, but allowed it through.

This is the PRL-8-53 thread, they just haven't changed the title, I reported it yesterday or so and asked them to.

The patent does say that the oral dosages range from 0.01-4mg/kg (most preferable 0.05-1.2mg/kg), the study used no such dosages however (they used 5mg for all).

Oral LD50 860mg/kg in mice (about 70mg/kg for a 60 kg adult)
Blood pressure in dogs was not affected in a dose of 8mg/kg (4 mg/kg in humans)
Reduced motor activity in mice and rats by 50% in 160mg/kg (13 mg/kg and 26 mg/kg in human dosage)
Does not potentiate amphetamine nor have any stimulatory effects like amphetamine.
No side-effects has been reported.
Reproduction has also been studied, there was no case of malformation of any form for two generations of offsprings.
Cancer risk is unknown, but if no malformation happened with two generations born on it, I doubt there is a cancer risk.

Hi Everyone,

Apologies for the delay in posting an update here... unfortunately due to work commitments I have had absolutely zero free time over the past week ;)

I have checked with the lab regarding how the CUSTOM SYNTHESIS is progressing and here is their reply, which is self-explanatory:

PRL preparation is at full stop because my supplier is stalling delivery of sodium cyanide which is needed at next step of preparation.

I have now about 250g of pure:

And good practice called for ordering sodium cyanide in excess (500g) for which there is much trouble/paperwork/scanning of documents/etc.

I need this poison to make next intermediate - 3-(carbomethoxy)phenylacetonitrile:

And before you ask - there will be absolutely no cyanides after this reaction in the product.

So there would be some delay, I think this poisonous reagent should arrive next week at my lab and there are 3 more steps to finished product plus final purification.


So good news and bad news everyone... The good news is that the CUSTOM SYNTHESIS is well underway... The bad news is it is going to take a bit longer than originally estimated to complete for the above reason ;)

I will be sure to post further UPDATES here as and when :)

EDIT: I should add that it is quite common for TOXIC CHEMICALS, such as SODIUM CYANIDE, to be used as REAGENTS within the CUSTOM SYNTHESIS PROCESS; wherein, what is important it that the lab used is sufficiently skilled and reputable such that no traces of said TOXIC CHEMICALS remain in the FINAL PRODUCT (which can always be confirmed via independent third party analysis) ;)

I asked them what was the cause of the delay and they confirmed that they have in fact carried out the synthesis twice as the first batch did not pass their strict quality control protocol, so they discarded it. They have informed me that are confident the second batch should do so. It is apparently a difficult substance to synthesize and because SODIUM CYANIDE is used as a reagent it is obviously extremely important that the synthesis and purification processes are carried out properly and thoroughly. So hopefully not long now! :)

Just wanted to let everyone know that I know of a trusted lab that finished synthesizing PRL-8-53, NSI-189 and IDRA-21... If anyone is interested in any of these compounds please email me at founder@gafoundations.org with how many grams you want... if I get enough people who are interested in PRL & IDRA I'll send out invoices.
