Curing brain damage is not nootropic potential for healthy people.

And I doubt anyone will ever (or well, in the coming decades) play around with fetal stem cells at home.


I'd rather not turn this into an NSI-566 thread (that time will come... maybe), but I could still see it being a nootropic; NSI-189, although being studied for nootropic purposes (according to NeuralStem), was originally designed for depression and brain disorders. Most nootropics are originally designed for these purposes (such as Coluracetam, for instance).

Does anyone have any idea as to how I can quantify my experience with NSI to the best of my abilities? I have severe HPPD and obviously if there is any reduction in my symptoms it would be evident, yet anyone can say, "(insert substance here) makes me feel great!" I was thinking about doing the typical cambridge brain sciences along with DNB and maybe Lumosity, but if I could find some way to get a rough approximation of my IQ (or even take a true IQ test), I'd do it. I may be getting SPECT scans and EEG's done at this real nifty neuroscience facility, but I don't know if I'd be able to afford a second round of scans simply to quantify this experiment.

I fully agree, but there are a lot more not working in healthy people than there are working, which was my point.

I don't plan on doing it for a few months though.

Does anyone know how long before it expires?

Should I keep it in the fridge or something?

I agree. I believe the implantation surgery costs something like $100,000 and will definitely be something that will only be used for approved conditions in the USA. Here is a site with pictures of the scarring aftermath of the NSI-566:

3/10/14 Update from Neuralstem on NSI-189 - Early review of Phase 1b data is encouraging, Neuralstem is expecting to start Phase II trial of NSI-189 later this year:

"2013 also brought significant progress in our neurogenic small molecule program. The company completed dosing of Phase Ib patients for NSI-189, in major depressive disorder (MDD), in August. A comprehensive review of the data is currently underway. Measurements include tolerability, pharmacokinetics, and pharmacodynamic effects including hippocampal volume by MRI, BDNF/CRH/other factors in blood, urine biomarkers, cortisol in saliva, qEEG, MADRS, NGH Depression Questionnaire, and Columbia-Suicidal Severity Rating Scale. While this data analysis will take us through the second quarter of 2014, the early review is encouraging. It looks like NSI-189 is taken orally in the study - is there any reason that we prefer sublingually?

Sublingual usually gives better bioavailability (0% should theoretically be destroyed as it can be in the stomach acid).

25mg twice daily, taken in a capsule. I will have to try it sublingually but I doubt this in itself will change the outcome

Interesting you say that, I have some minor nausea, and a lot of bloating/intestinal discomfort. Don't know if it's the NSI or the flouride shield I'm taking (recently upped the dosage quite a bit).

I am currently taking 40mg x2 (day 5) sublingually. I occasionally get anxious in the middle of the day as well. It feels like a small pressure inside my head like an annoying tick. It does go away after a little while.

No nausea for me, I have noticed a good sized chunk decrease in stamina and endurance though (I train every day for 2-3 hours).

Long time lurker, First time post.
Received nsi 4 days ago:

Not getting a lot from it up to now seems to make me very unmotivated and a bit groggy, A little dazed at times, A bit of a headache today not a big deal.
Dossing at 40mg sublingual (BID) Getting a lot of extra salivation, Makes it very hard to take this way and there's always quite a bit of powder left which I have no choice but to swallow, Doesn’t seem to be very water soluble.

Just wonder if this is the best way to take it or just to take it orally like the trials, They presumably would have already accounted for the amount lost through digestion when deciding on the 40mg dosage, Which would mean taking it sublingual 40mg would be over kill.

Seems to have a similar smell to PRL-8-53 just not as strong.

Supplements:
uridine 250mg (sublingual)
fish oil 3g
CDP 400mg
multi
b vits

I take 20mg in a capsule with food.

Sublingual is making me high and it rushes in fast. Eating it makes the effect more gradually. I can feel that it works for about nine hours. After 7 quarters till two hours I got a slight headache and a high feeling and a bit later energized. When I take it I don't want to go to the gym. After three hours I can't wait to go. After four hours I get muscle pain in the neck and shoulders and can't focus as easily anymore. This only appears when I'm hightening the dose and it's from day three. After a week it calms down. Sometimes it gives some anxiety.

I take also 7 grams of fresh turmeric root with pepper and olive oil. If I have to believe some research it has also neuralgenesis properties and should even work as a AD.

I have no clue if these interfere with each other or if they cooperate.

sounds very similar to 7,8dhf if i take a bit too much i get a pressure headache nausea and foggy head. I have a feeling increased bdnf-like activity has a therapeutic limit no matter how potent the substance.

I have about 50 to 100mg left from the first batch and I will do side by side comparison when my new vlk nsi-189 arrives. Hope this helps.

I manage a chemistry lab that has a DSC, FTIR, and pyGCMS we could use for purity testing.

I also have this website called https://quantimo.do/ that I made to integrate self-tracking data regarding my mood and any other variable to mine if for correlations and hidden influences on mood. My goal is crowdsourcing clinical research by anonymizing and collecting the data from all these underground experiments.Otherwise, at best all we have is thousands of subjective forum posts that can't be aggregated to gain greater insight into effectiveness rates for specific symptomatalogical and genetic profiles.

Also, we can more rapidly accumulate safety and efficacy data which could be used to pressure regulatory agencies to reduce restrictions. Please PM or if you'd like to know more. Thanks!

The BDNF increasing agents appear to as well effect dopaminergic activity, primarily.
BDNF agents are indeed something of worth, but will generally be of limited efficacy.
Many common agents that have been well studied and in common use for many years, such as selegiline, provide significant BDNF enhancement, but are not nearly panaceas ;)

There is perhaps nothing too novel here, or within other 'newer' BDNF enhancers.
Again, they certainly have worth. As with any use of any substance YMMV ;)

Right! Thanx. I didn't know that selegiline was also a BDNF enhancer - actually I just looked the word up in the Wikipedia, it was unknown to me... So my knowledge grows, hopefully with the size of my hippo. :)
