The injections are actually "oral". The mice received ip injections which means it is injected into their stomach.
But i think it is probably impossible to overdose with 5 mg for a human.


I don't think we should forget about the potential of such a supplement being taken accidentally by a kid when they see daddy doing to be smarter. I'm thinking we should have it so you take more, smaller capsules throughout the day. There is a danger potential here if a regular supplement is developed and we need to make sure we do this responsibly so we don't attract any negative attention.

The first is C16 and the other is C-19. Are they similar? The downside of the C16 is that it is only 90% pure but the C-19 is 98% pure... The ISRIB does sound like a more stable and easy to use product. I have not found anyone who advertises its synthesis for sale.

How does it compare to the stuff used in the study? Maybe it was only 90% also? I wish pubmed was still publishing full papers...

I don't have enough data but I would think that 90% is not the highest grade possible. The 90% proof C16 only speaks of the product from santa cruz biotechnology, inc. Why they don't bring it to 98+% I do no know. Perhaps a phone call might prove they could and would for a higher price. I would be interested to know if anyone has any information on C19. I have no idea how it compares. If I get some more time I will dig deeper.

From what I have been reading ISRIB sounds like a better candidate that does about the same thing. I will wait to hear from that group before jumping head first into the C16's car. :)

Depends on how big your doses are (which also depends on you're bodyweight), but maybe around 50 doses or something.
Now that I am thinking about it, I think I remembered paying around 150 USD for the 5 mg. The extra cost is the VAT and transportation and some other bureautic stuff.

I'd be getting it in the states, so no red tape/tax for me. Did anyone ever find something better than DMSO for it? Emu oil is a valid alternative?

If you're going to get it, than I would advise DMSO. Because it is known not to interact with DMSO. The compound might be so unstable for instance that it might react with the Emu oil before it has the chance to do its work. Wich is also the reason why you first have to degas the DMSO with an ultrasonic cleaner. The problem with c16 is that you don't know how unstable it might be (could oxidize or polymerize).
If you're going to weigh it (probably also best with microgram scale), then it will probably be best to do it in a glovebox.
Anyway Googoltarian knows more about this than me. It would be best if you contacted him.

Glovebox? Wasn't someone looking into testing a sample for stability? How can we determine a good delivery method? I'm really not comfortable with DMSO... How could we test for it in a solution?

I have some good news regarding C16.

I have spoken to Googoltarian and we arranged that I will receive the C16 in the middle/end of august. I know the entire “C16-trial” has been a bit of an ordeal and keeps dragging on. First the courier woudn't transport it. And thereafter, everytime I had the chance to receive the compound the past month, Googoltarian was too busy and right now my schedule is filling up again too.
But I think we all should be glad that we are working on this, and believe me that we are working hard on it, because clinical trials of this sort (with dephosphorylation of eif2alfa) are still years away.

If this time everything goes well my first results will be in by the end of next month/the beginning of september. So that is a date you can all look forward to. So if everybody could be patient for just a little bit more in the meantime, that would be great :-D . I have been wanting to try this since the first reports about pkr inhibition almost 2 years ago and believe me I am not going to back out! Doing this is also not as easy as it looks. Preparing a vial for usage is not that simple since it requires some instruments such as an ultrasonic cleaner, a microgram scale,... The compound might polymerize before it can do it's job, so I don't want to screw it up.

About the ISRIB group buy:
I think we should do everything to get this around! Even if the pkr inhibitor doen't work, because the ISRIB works in a different way than C16. ISRIB prevents phosphorylation of eif2alfa by inhibiting one of the four other kinases (PERK I think). Or it might just be possible that the inhibitor is ineffective in humans (human and mouse pkr are slightly different). Or the C16 interacts different in humans than in mice.
So i say we push through with the ISRIB group buy even if the C16 doen't work. And if we need another supplier, so be it. I am willing to help find a new custom synthesis lab. I am even willing to organize an new group buy, if I am able to do that (I don't think normal individuals can order something from a custom synthesis lab without proper documentation). I am starting to get interested in these group buys, since they allow us to reach chemicals we normally coudn't.

Organizing these group buys are great, but I think they are not that easy (there will be setbacks). But if we all work together and are a bit patient, than I think we can achieve something quite impressive.

From what I read on another thread JPC16 hasn't received the C16 yet. I hope the shipping company is keeping it cool.

Since everybody is so curious: I still don't have any feedback about C16 :sad:. Because I still don't have it yet. I am still in the middle of trying to get the compound to me so I can start the trials.
The reason why it takes so long is that Googoltarian seems to be really busy all the time because he sometimes is offline for 2 weeks. And the last time he wanted to send it to me he suddenly was out of town. But I refuse to let these setbacks hold me down! I will get my hands on C16 even if it is the last thing I do. I have been researching and working on this for a very long time and I won't stop now. I will get this done, trust me. It's just a matter of time.

But if it's any consolation, when I do finally receive the C16 (and I will ! :-D) then I will be able to execute the trial pretty fast. Since the academic year started for me which means I can “easily” get my hands on a microgram scale to prepare a dosage. But on the other hand, I will also be a bit busier.

For now, I can't give you guys an estimate when things will get around. But I will keep you posted.
