From the video, it shows fasting for 5 days really beneficial like lower IGF-1 and Insulin level. And it also mentioned fasting will remove those bad and old cells, and create new young cells after fasting over.
So, perhaps those who take NR has no effects should try the combo of fasting and NR. Fasting remove old, bad cells and NR increase NAD+. Both seems to work together perfectly. I guess one will become a lot more refresh and young using the combo of fasting and NR then either alone.

Well, I think the benefits of fasting are well-demonstrated, especially in regards to the immune system and slashing inflammation.
But I'm not sure your experiment makes sense. How would you tell?
I'm not saying it's bad idea in terms of personal health, I just saying it confounds the results.

By simple logic....NAD+ like nuts and bolts, plier, wrench, ...tools. Fasting like the body now become a mechanic. With plenty of nuts and bolts, so the mechanic easily repair the machine (our body). Does it sound logic?
I mean after killing bad and old cells, supplying the body with plenty of NAD+ to new and young cells really will much better than the young and new cells with not much of NAD+ (fuel). Please prove me wrong by testing. I would like to try but fasting is really really difficult for me.

I have never been able to lose weight before until I started increasing Niagen dose to 375 mg. now I can see benefit from exercise and lose 8 lbs in two months.

Hi all, long-time lurker, first post. Been following this thread quite a while and want to say I appreciate the many people who have posted their personal experiences here. I started NR about 5 months ago.
Background: Very healthy moderately active 64, other than borderline BP and would like to lose a few pounds. On supplements for years (i.e. ubiquinol, omega-3s, B, C, D, mag threonate, curcumin, taurine, etc.).
In general, I don't take supps to "feel something." My goal is long term health and wellbeing.
Started NR (HPN) at 125 in the a.m. for a couple days then doubled it to 250. Within a few days I began to feel a marked lift in my energy level, like there was a pool of reserve stamina. It was an amazing new sensation compared to how I felt a week before, but I wouldn't call it a buzz. It wasn't brain related, it was physical. I felt lighter, found I could get up and move without stiffness, exercise as much as I felt like, push myself and recover without dragging or aching the next day. Even though I thought I had felt just fine before starting NR, I guess it sort of lifted a weight I didn't realize was there. The bouncy sensation lasted for about a month before it stopped being something I really noticed. It turned into a new baseline.
After a few weeks I increased my dose to 375 in the a.m., and that's where I've stayed.
Other observations: my skin has gotten softer and smoother all over; age or sun spots I had on my hands for well over 10 years from too much beach time have faded and seem to be disappearing. Not sure if it's related to that, but although I've been told I look young for my age since my 50s, it's becoming a more predictable reaction lately. Other things: borderline sleep issues I was starting to get cleared up and I'm sleeping very soundly again. Also, very big deal: I have flat feet and a bunion on the right foot which has always caused foot pain and aching after walking a lot or being on my feet too much. After 20 years, no more foot pain! I still have the bunion and my foot is still flat--but no more hobbling around the day after a hike!
I haven't noticed any particular weight loss from it and haven't been trying, really. Just enjoying being a lot more active without the downsides.
I plan to keep taking NR and hope the benefits continue. To those who were hoping for a noticeable "buzz" from it-- it didn't work that way for me. I agree with others who say if you are still young(ish), healthy and fit, improvements at the cellular level might be too subtle to notice as a physical or mental sensation. Doesn't mean they aren't happening, though.

Could you share your daily dosage, age, health status, and how long have you take NR? Do you take NR in the morning with plenty of water and short gap before taking your meal?

500mg in the morning without food, plenty of water for sure. 500mg again at night maybe an hour after food. I'm 41, healthy, regular exerciser. Still have some weight to lose but on NR it is just as hard (if not harder) to shed those pounds.

Yeah it's $4 a day, not the end of the world. I don't need to have faith in Sinclair, it's a decision I made based on the science available today. I believe it's a good bet based on the risk / reward ratio. Just because it doesn't help with fat loss, or other obvious effects, that doesn't mean it doesn't have positive effects.

A 45 year old male lost 10 lbs in one month. People seeing good effects are likely not in perfect shape physically or older.

you lose weight when you are in calories deficit, there is no going around that, i really dont see how NR can help with weight loss, unless it works as appetite suppressor.

I guess your assumption is that it's all about calorie consumption.
That's not logical. Even if you believe that a calorie is a calorie (pretty much disproven at this point), there are two sides to calorie balance: Calories In, and Calories Burned*.
- If you eat the same amount and NR speeds your metabolism, you'd lose weight.
- If you eat the same amount and NR swaps muscle for fat, you'd lose weight.
- If you eat the same amount and NR makes you more active, you'd lose weight.
- If you eat the same amount and NR 'makes you younger', you'd probably lose weight; at the same caloric intake, people tend to gain weight as they get older.
- If you eat the same amount and NR readjusts your hormonal balance in the right direction, you'd lose weight.
Now, before you jump all over me, I'm not claiming NR makes you lose weight. I don't know, and don't have an opinion. But I do know there many many possible ways it could other than appetite suppression!
*Calories Burned isn't the only use, either. There are Calories Excreted, and Structural Calories (for example, amino acids incorporated into muscle). Calories consumed that go down those paths can't be stored as fat.

That's right, the other factor is calories absorbed, it may be the case that nr helps or hinders adsorption of carbs. Sharing my sister experience UPDATE:
Week1
After taking NR 275mg/day for 1 week, now her constipation seems to improve. Really unexpected though. I mean I really can't relate the connection between constipation and NAD+. I was expecting she said something like feel more energetic, feel better sleep, etc....but she said constipation improve and getting better! of course, she doesn't have problem sleeping and constipation is her only main concern for long time...still...
Week2
Feel more stamina after gym, not feeling like tired or soreness after gym.
Age: 57, Female, Healthy, gym 1-2 times a week. pre-diabetic, BP on high side (still normal). long time constipation problem.

I wonder if anyone has done their uBiome before and after NR and if there was any difference. According to Art Ayers (coolinginflamation.Com) constipation is primarily a lack of bacteria in the bowel. IF NR has an pre or pro biotic effect on the microbiome, as many things do then that might explain the effect.
I would also say my constipation was improved and has worsened since being off NR but there are other factors
I believe NR has no effect on pro biotic. However, I believe (pure guessing) NR does improve liver, kidney, heart, ... a bit in general, and thus overall your body feel better and more energetic, that translates to better and helping in constipation too. All organs all inter-connected. The human body is a small universe, each organ somehow depends on other organ and so on, it is a cycle. All organs need NAD+ as fuel, and the fact that NR increase NAD+, so in general, all organs now got more fuel and function better.

Wow, you clearly have access to sources I don't have. But what's your point, and why are you posting this "Personal Experience" thread?
Can you please share with us your evidence as to which people were paid to endorse this product, and by who?
Or is this one of those "I'm saying this and therefore it is correct unless someone can prove it isn't" things?
The fact that people are increasingly taking that approach doesn't mean it's okay. It's a cheap and intellectually dishonest tactic.
