I know there is a already famous NSI-189 thread, but it is filled with too many incomplete experiences and also not everyone using it is using it for depression/PSD/Anxiety it would be wonderful if people who used or are currently used NSI-189 would share their stories on how it helped their depression to help others out and maybe give them some hope.

Some people may not know enough or be comfortable enough to purchase a drug online and test it on themselves that takes a lot of guts imo.  But also because it won't be officially released for quite some time, I know people come on here trying to find good outcomes form other people who have tried it.  Finally depressed people always need hope.

Hope is related to placebo..and last time I checked placebo and antidepressants don't differ much in efficacy.
Also, nsi-189 is life changing for me. I'm still feeling the effects strongly nearly 5 weeks later.

I am very interested myself as I have chronic low level depression quite possibly related to inflamation from leaky gut syndrome, I am much better now, following a special diet, but I am still having some symptoms of mild brain fog and mild depression that get better with NSI-189.

 

I am interested to give some NSI-189 to a past girlfriend and very good friend, and looking to see if anyone combined a SSRI (uses paroxetine for two years now) with NSI-189?

 

My experience answering a pm.

"For me, using 20mg gives me a calm feeling, 40mg a good mood boost and an increased feeling of confidence, I have very "confident ideas", but I do not feel as confident, NSI helps with that. At around 60mg I can get a little speedy and anxious, depending on circumstances also. Around 40mg its best for me, and I do not use more than this, at one time. 

 

What I find interesting is that I get some instinctive reactions, some "deep emotions" that I usually do not have as I am usually very cool headed. What I mean is that I am, and feel more emotionally and "instinct" directed than my extremely rational self. I have also seen a mild increase in my information processing capacity, and I am having better memory for trivial facts."

Hi, this is my 1st post on Longecity but I've been a lurker for approx 5 years.
I've found this site very informative and there are a few regular posters/members that over the years I've learnt a great deal from.....I've also learnt to separate the wheat from the chaff if you get my drift.
A little bit about me: I'm male, the wrong side of 50. I've worked 30 years full time as a Paramedic working (12 hr rotating shifts). For most of my adult life I've battled with depression and early this year I was diagnosed with PTSD and had EMDR therapy which seemed partially successful. I'm relatively psysically well, bit stiff in the morning (not in a good way) and have Gilbert's syndrome which sometimes increases my fatigue.

Today my order of NSI-189 arrived and I can't believe it but my mood seems brighter and my senses seem heightened at the end of my 1st day. Of course I cannot be sure it's not a placebo effect. I'd like to add though I've tried many supplements/nootropics over the years with high hopes and never felt like this.
I realise this is extremely early days and only time will tell but I feel this is the real deal. Btw, I'm taking the tablet phosphate form 20mg bd.
I just felt the need to share this with Longecity and will keep the forum updated.

I felt the effects of NSI from the first day on as well. Its been the best anti-depressant/noot that I have used.  Between it and TRT + B12 shots, the combo has taken care of most of the issues I was grappling with for many years, many of which were exacerbated in the last 4 due to familial and financial stress.

 

I think high cortisol and low Test were at the core of most of my issues. Correcting that ratio along with the sort of receptor repair work that the research on NSI seems to indicate have made life a hell of a lot better. Anhedonia, depression, low/no libido, chronic fatigue, suicidal ideations, emotional numbness and irritability are all now much removed from my daily thought processes. It's been a big relief as this has been the butterfly I've been chasing for years with little success. I really hope NSI can be used long term or that its effects last long term.

I've used it twice, for a couple of months each time.   I will also use it again soon.  I too, really wonder if it would be synergistic with an SSRI (and CBT).  I found it slightly raised my anxiety levels after an initial honeymoon period, where I immediately felt a deep, profound sense of connectedness. The closest thing I can compare that feeling to is the afterglow of a positive LSD trip a month or two later.  

 

I do think it has had positive effects on my mood that seem permanent, but after such a profound initial reaction, I made my usual mistake of believing I needed to increase the dose to chase the same experience.

 

Now I believe it's an extremely powerful tool, with minimal (none in my case) side effects, to fight depression; but, I'd love to run it for an extended period of time, in conjunction with CBT/meditation and possibly an SSRI, just to see how powerful it could be.

I've actually used the two together and have a couple bad reactions.  In both cases, I was using Tianeptine powder and most likely used in the 40-50mg range v. the 12.5 dose.  Both times I was driving long distances and was also taking a relatively low dose of Modafinil (for me).  In any event, both times I immediately felt a loss of equilibrium, and a sense of dissociation.  In both cases, thanks to Tianeptine's short duration, I felt better rather quickly.

 

In general, while I believe Tianeptine has a great deal of potential, unless a time-release version is formulated, the temptation to re-dose and re-dose is too great.  I find myself far over the 12.5 3x per day dose within a couple days, and soon am extremely constipated.

 

I thought the Tianeptine Sulfate version might be a solution, but in my case it was not nearly as effective as the sodium version.

 

BTW  I've taken Prozac, Zoloft, Lexapro, as well as Effexor, and Pristiq.

 

Of those medications, Lexapro was the "cleanest," and Pristiq the most effective.  Thankfully, I had ZERO withdrawals upon cessation of all of the above.  I'm very lucky that way.

There is also just something about SSRi's that allows you to unburden, for lack of a better word, with a good therapist that I didn't feel with Tianeptine.   Again, in my experience, Tianeptine was like a better, albeit short lasting, version of Wellbutrin...without the crankiness and mood disturbances I had while taking Wellbutrin.

 

Now NSi-189 was a different beast entirely.

I'm about to order some NSI-189, but quite nervous ordering online a drug that hasn't been fully tested. That being said, i am intrigued by it's mechanism of action - increasing hippocampal volume. 

 

Wondering if people have had any negative reactions? Or are they mostly positive?

 

I'm interested in the freebase, not the phosphate. Is there an issue with using one or the other?

 

What would the dosing be - how much, how often?? Want it to maximize hippocampus volume, as mine was quite small according to a recent MRI :(

Hi all, I meant to get this posted over a month ago, but I'm so busy now with research activities on interests that intrigues me, I just felt like I don't have time, but I see I had to make the time right now.
I put up a post, I believe in Feb, this year, almost begging for some knowledge someone had that I didn't on what happened to me. It's in the mental health section. Anyway I messaged a member on here, got a bit of NSI-189 to try. The results for me have been outstanding beyond what I could have ever hoped for. Surely one may wonder, well maybe time that passed helped and I don't know but beg to differ.
When I finished all I had, which was about the first of May, I can't begin to put into words the changes I have now, in my thinking, my outlook, my Intellect, I have realized so much like never before, I feel as though something switched on in my brain, I'm trying to word this best as I can, lol. But I feel so mentally strong now that I can take anything, it's just life, take it and move on. It has opened up interests that I never would have believed was in me, So those interests now have me seeking and searching proven relavent info on it and I just can't take enough of it in fast enough, lol. Stuff as, cognitive psychology and the whole human thought process, why we think the things we do, how evolution and natural selection brought our species into this, the whole universe and the realitization of the origins of life on earth. There are categories that go with each of these that I keep finding that is so amazing. Like I want to know all I can about it. For me this has been my life changer and to read my initial outcry for help, you'll see why.

Wondering where you get yours from and whether you use the freebase or the salts? I just ordered the freebase, but concerned that it's not What NeuralStem has been testing.

 

Also, what dosage are you using?

Hi. I ordered from a member on here as another member gave me their profile name. The name is StrangeLove, very reputable. You pay through paypal and it's sent to you via USPS. I don't know the difference in free base vs salts. This I got was a fine powder, with a very very slight yellow tone. I started out with 25 milligrams as instructed by StrangeLove and kept it at that for maybe 4 days then increased to 30 for a week, then to 40 and stayed on that for about 3 weeks, then went to 45, sometimes 50 but no set time frame for the 45, 50 I just finished taking all I had left at this point, I was meaning to stay at 45 but I figured the few extra wouldn't hurt, I took one dose a day. At first I had anxiety from hell, constantly but I read others did too, don't know what's up with that and after the first 2, 3 weeks into it that's when I noticed the change. I kept a log and wrote down my feeling every day. The change was so "there" it got my attention to where I had to stop my thoughts and reflect on it.

Worked like a charm for my burnout-depression - after but a few days I regained tons of energy, mood was improved, and overall my cognition improved SUBSTANTIALLY.

 

However, the effects seemed to plane out at around the 3-4 week mark, after that it had diminishing dividends and eventually it just made me really, really tired.

 

 

Overall I am happy with the results I've had on it though, and I've actually come back and done two more cycles, with varying time-frames and results.

I also found that phosphate was substantially more effective than freebase, even when the freebase was taken sublingually. IN closing, I feel that it seems to work the best for fatigued, or anhedonic depression, while for anxious depression it really doesn't seem to work as well.

Probably because anxious depression is really a different disease, I guess. (we know regular anxiety certainly is - completely different mechanism - it just so happens that several drugs that work for one, work for the other - but DEFINITIVELY not all of them - some are really bad for one or the other)

 

I was also sensitive to the dosing - when I was the most burnt out, then I could take 40 mg with good results, but pretty soon this turned into too high of a dosage, and NSI started inducing anxiety and odd, panic-like, mood-swings - I also became a bit too emotional (sort of the same thing, I suppose), at 40 mg. So I dropped the dosage down to 20 mg and got much better results.

 

It should be noted, that I actually had SOME positive results on depression and burnout at as low of a dosage as 10 mg...! : O And I'm not the only - the 2 x 20 mg dosing-scheme is correct for most people, but it's not universal, just like with any drug, there are outliers - as such, if you/anyone feel anxious after taking the drug, then don't hesitate to LOWER your dosage, ever so slightly.

Why would the phosphate be more effective than the freebase if the only difference should be pharmacokinetics?

Anyone has experience mixing NSI with alcohol?

 

Alcohol is a pro-depressant and neuroatrophic - the opposite of NSI-189.

 

No one who's depressed should ever drink - I suggest you simply quit drinking.

(unless you're addicted, then I would suggest you seek help - and possibly add CERC-501 or Naltrexone + Buprenorphine to your regimen)
 

I have no personal experience of the combo btw, since I am, and always will be, a teetotaller.
