Great links and a very personal insight to your main motive regarding NR, thank you for this, Bryan.

I want to give you guys something back for all the things I've learned through this forum and it's even NAD+ related.

I started smoking pot relatively young and stopped it one and a half years ago, but I still suffered from a light bipolar disorder.

Then I stumbled over this article:

Why Aging People Become Depressed, Fatigued, and Overweight

http://www.lifeexten...rweight/Page-01

http://www.lifeexten...rweight/Page-02

So recently I stopped Nicotine as well by strengthening my will through vegan Tryptophan supplementation (->Serotonin, Melatonin, Niacin).

Interestingly, my mood became very stable, less thoughts about stupid humanity and my circadian rhythm improved significantly.

Previously I stayed up till 03:00 am almost every night and now I get tired when the sun goes down :-D

I can highly recommend it to support your soberness or to even become a better human? (dramatic)

It's the least common essential amino acid in our food. I think there's more to it than modern science knows to date...a evolutionary reward system anyone?

This site could be interesting to you guys aswell, it's basically an overview of natural active substances:

NAD+ is synthesised via two major pathways in humans; the NAD de novo biosynthesis and NAD salvage pathways...the de novo pathway starts with L-tryptophan...humans stricken with pellagra, a nicotinamide (niacin) deficiency disease, recovered after the addition of tryptophan...

http://humancyc.org/...ject=NADSYN-PWY

NAD+ synthesis pathways are categorized into either de novo pathways, which start with the amino acid aspartate or tryptophan, or salvage pathways, which start with nicotinamide (NAM) or nicotinic acid (NA)

http://journals.plos...al.pgen.1001105

In all vertebrates, NAD can be synthesized by...de novo synthesis from tryptophan...For the de novo pathway to be completed it is necessary to have adequate riboflavin (vitamin B2), pyridoxyl phosphate (vitamin B6), and ascorbate (vitamin C).

http://www.ncbi.nlm....les/PMC2651433/

With increasing age...profiling of blood serum displayed a marked decrease in tryptophan concentration

Thank you for the links, Logic :)

I should have Vitamin B2, B6 and C covered (VEG1 Supplement + fruits)

An intake of 60 mg Tryptophan result in 1 mg Niacin metabolite

http://www.ncbi.nlm....pubmed/15895512

So 3500 mg Tryptophan result in 58 mg Niacin if it's converted linearly

I've been told that tryptophan increases quinolinic acid,a marker for Neuro degeneration.

quinolinate is an intermediate of de novo biosynthesis pathway

ChromaDex Initiates Second Human Clinical Study on NIAGEN® -The World’s First and Only Commercially Available Form of Nicotinamide Riboside (NR)

Broader Study Will Build Upon the Successful Results of the First Trial Which Demonstrated NIAGEN® is a Safe, Effective NAD+ Precursor in Humans -

IRVINE, Calif., March 10, 2016 (GLOBE NEWSWIRE) -- ChromaDex Corp. (OTCQX:CDXC), an innovator of proprietary health, wellness and nutritional ingredients, that creates science-based solutions to dietary supplement, food and beverage, skin care, sports nutrition, and pharmaceutical products, announced today the initiation of the second human clinical trial for its patented ingredient, NIAGEN® (Nicotinamide Riboside - NR).

The second human clinical trial will build upon the results of the Company’s first human clinical trial announced in February 2015 which demonstrated that a single oral dose of NIAGEN® is a safe, effective nicotinamide adenine dinucleotide (NAD+) precursor in humans.  The results of that study constituted a significant milestone as it showed for the first time an increase in NAD+ in humans has been demonstrated through NR supplementation.

The second human clinical trial will be a randomized, double-blind, placebo controlled parallel study in 140 healthy adults ranging in age from 40-60, which will examine the benefits of NIAGEN® if taken daily for eight consecutive weeks.  The outcome will provide key information on the effective dose range of NIAGEN® to increase the mitochondrial co-enzyme NAD+ and NAD+ metabolite concentrations in the body.

Additional endpoints to be investigated include: C-reactive protein, total cholesterol, LDL, HDL and triglycerides and amino acid panels, NAD+ levels in muscle, mitochondrial biomarkers, and the effects of NR on Resting Metabolic Rate.

Frank Jaksch Jr., founder and CEO of ChromaDex, commented “Given the success of the first human trial of NIAGEN®, we have been eager to generate additional human data demonstrating NR is an effective precursor to NAD+.”

Maybe the results of the second test will actually get published.......the results from the first never were, which doesn't necessarily bode well.
That being said I still take 500 mg a day in divided doses

Unfortunately I have to revise a recommendation, highly dosed Tryptophan or its metabolites can cause Eosinophilia-myalgia syndrome (EMS) in some individuals.

Post-epidemic eosinophilia myalgia syndrome associated with L-Tryptophan

Eosinophilia-myalgia syndrome induced by excessive L-tryptophan intake from cashew nuts

It was good while it lasted...I'll stick to Niacin.

Form 8-K for CHROMADEX CORP.

https://biz.yahoo.co...11/cdxc8-k.html

Item 1.01. Entry into a Material Definitive Agreement

On March 11, 2016, ChromaDex Corporation, a Delaware corporation (the "Company") entered into a Securities Purchase Agreement (the "Purchase Agreement") with an existing stockholder pursuant to which the Company agreed to sell in a registered direct offering a total of 384,615 units (the "Units") at a purchase price of $1.30 per Unit, with each Unit consisting of one share ('Shares") of the Company's common stock, par value of $0.001 per share ("Common Stock"), and one unregistered warrant (the "Warrants") to purchase one half of a share of Common Stock (the "Warrant Shares"). The Common Stock and the Warrants are immediately separable and will be issued separately. The Company has not engaged an underwriter or placement agent in connection with this offering.

I don't normally concentrate on the company business but here we can see they are in the process of raising half a million dollars to further some objective ahead.

If I recall well they stated that they may raise some cash as they plan to uplist exchange. Likely not related to research.

Well you seen it very fastly after i published it.. Impressive Bryan. Im working on it only within my free time and I think on the long run it would be interesting to have a summary from this thread into one blog with different title for each topic for instance the sublingual part, the mechanism etc.. On the forum its good to discuss and debate but very hard for some to use the googlesearch bar from this forum sometimes. And its unfortunate because the work / research done here is very good.

Science News
Mitochondrial metabolism linked to acute kidney injury

PGC1 alpha works through NAD 'aging molecule' to guard against stress; research offers new therapeutic target for acute kidney injury

https://www.scienced...60316151353.htm

Here we go . . . I thought this was appropriate to mention. I think this NAD+ story keeps getting stronger and stronger.

kidney.png

"Within the kidney there is abundant ATP and mitochondria to carry out this essential chemical function." said Parikh. "We hypothesized that mitochondria might also be serving as an essential and early target for the various risk factors leading to kidney injury."

"One of the most robust reactions was a local deficiency of niacinamide [an analog of vitamin B3] within the injured kidney," said Parikh. Vitamin B3 serves as a precursor for NAD, the 'aging molecule' at the heart of energy metabolism. "Recent studies have suggested that NAD levels fall with aging or with diabetes, which are two of the major risk factors for kidney injury. That was when a lightbulb went on."

To determine if increasing the level of NAD would accelerate recovery from kidney injury, the researchers administered extremely large quantities of vitamin B3 nutritional supplements -- the human equivalent of hundreds of tablets -to the genetically impaired mice.

"We found that, following administration of the supplements, the kidneys were able to resist both toxic and ischemic stress," said Parikh. "Normal doses of nutritional supplements are probably not going to cure kidney injury, but these experiments provide a proof-of-concept that metabolic manipulation of NAD might be a new avenue for the development of future therapies.

"Our work draws some new connections between PGC1 alpha and the NAD molecule, and their effects on metabolism," added Parikh. "If indeed NAD levels fall with aging, the kidney may be a new window to thinking about how we study aging overall and may take us outside the world of acute injury to more broadly consider how the body deals with environmental stressors throughout life."
