Great links and a very personal insight to your main motive regarding NR, thank you for this, Bryan.

 

I want to give you guys something back for all the things I've learned through this forum and it's even NAD+ related.

 

I started smoking pot relatively young and stopped it one and a half years ago, but I still suffered from a light bipolar disorder.

 

Then I stumbled over this article:

Interestingly, my mood became very stable, less thoughts about stupid humanity and my circadian rhythm improved significantly.

Previously I stayed up till 03:00 am almost every night and now I get tired when the sun goes down :-D

 

I can highly recommend it to support your soberness or to even become a better human? (dramatic)

 

It's the least common essential amino acid in our food. I think there's more to it than modern science knows to date...a evolutionary reward system anyone?

I should have Vitamin B2, B6 and C covered (VEG1 Supplement + fruits)

 

An intake of 60 mg Tryptophan result in 1 mg Niacin metabolite

IRVINE, Calif., March 10, 2016 (GLOBE NEWSWIRE) -- ChromaDex Corp. (OTCQX:CDXC), an innovator of proprietary health, wellness and nutritional ingredients, that creates science-based solutions to dietary supplement, food and beverage, skin care, sports nutrition, and pharmaceutical products, announced today the initiation of the second human clinical trial for its patented ingredient, NIAGEN® (Nicotinamide Riboside - NR).   

The second human clinical trial will build upon the results of the Company’s first human clinical trial announced in February 2015 which demonstrated that a single oral dose of NIAGEN® is a safe, effective nicotinamide adenine dinucleotide (NAD+) precursor in humans.  The results of that study constituted a significant milestone as it showed for the first time an increase in NAD+ in humans has been demonstrated through NR supplementation.

Additional endpoints to be investigated include: C-reactive protein, total cholesterol, LDL, HDL and triglycerides and amino acid panels, NAD+ levels in muscle, mitochondrial biomarkers, and the effects of NR on Resting Metabolic Rate.

Maybe the results of the second test will actually get published.......the results from the first never were, which doesn't necessarily bode well.
That being said I still take 500 mg a day in divided doses

Just did a Whois the site belongs to Andre Thomas who we all know. Guys I see he needs some help, PM him and see what he needs rather than poke at him. Sometimes the French to English translations also need some adjustment.

Unfortunately I have to revise a recommendation, highly dosed Tryptophan or its metabolites can cause Eosinophilia-myalgia syndrome (EMS) in some individuals.

Well you seen it very fastly after i published it.. Impressive Bryan. Im working on it only within my free time and I think on the long run it would be interesting to have a summary from this thread into one blog with different title for each topic for instance the sublingual part, the mechanism etc.. On the forum its good to discuss and debate but very hard for some to use the googlesearch bar from this forum sometimes. And its unfortunate because the work / research done here is very good.

"Within the kidney there is abundant ATP and mitochondria to carry out this essential chemical function." said Parikh. "We hypothesized that mitochondria might also be serving as an essential and early target for the various risk factors leading to kidney injury." 

 

"One of the most robust reactions was a local deficiency of niacinamide [an analog of vitamin B3] within the injured kidney," said Parikh. Vitamin B3 serves as a precursor for NAD, the 'aging molecule' at the heart of energy metabolism. "Recent studies have suggested that NAD levels fall with aging or with diabetes, which are two of the major risk factors for kidney injury. That was when a lightbulb went on."

To determine if increasing the level of NAD would accelerate recovery from kidney injury, the researchers administered extremely large quantities of vitamin B3 nutritional supplements -- the human equivalent of hundreds of tablets -to the genetically impaired mice.

"We found that, following administration of the supplements, the kidneys were able to resist both toxic and ischemic stress," said Parikh. "Normal doses of nutritional supplements are probably not going to cure kidney injury, but these experiments provide a proof-of-concept that metabolic manipulation of NAD might be a new avenue for the development of future therapies.

"Our work draws some new connections between PGC1 alpha and the NAD molecule, and their effects on metabolism," added Parikh. "If indeed NAD levels fall with aging, the kidney may be a new window to thinking about how we study aging overall and may take us outside the world of acute injury to more broadly consider how the body deals with environmental stressors throughout life."
