I am in for 20g.

I do have concerns, but we do not know when (if) another group buy will happen. So I will buy it and hold on to it for a bit.

Do you guys believe the complications associated with the drug are decreased with the smaller dose? Im talking things like hyperglycemia, Peripheral Artery Disease, and the sudden death associated with prolonged QT interval.

Any input would be helpful

Good to have you on board Roydeman

I do plan to do group buys every 1-3 months and plan to post that the buy stays open.

ie: those that want just post a commitment and once there are enough commits to start collecting funds I will do so.

Minimum collective order is 200 grams and the next buy should be cheaper as we will probably have trust in the supplier and forgo testing, but that will be up to everyone.

The side effects were said to be non existent at the smaller doses of 150-300 mg IIRC Roydeman, but a stack to counteract those effects is a good idea.

Pterostilbene would be on my list as a guard against hyperglycemia and Peripheral Artery Disease, plus a host of other benefits.

http://www.pterostilbene.com/

I'm sure others will recomend the other things I don't remember off the top of my head too.

Cinnamon etc.

I also recomend taking a potassium and magnesium supp 1st.

Like this one perhaps, but I don't know the supplier's reputation:

http://www.gnc.com/G...ductId=16655316

Perhaps have BHT on hand:
it will upregulate CYP3A4 which clears Nilotinib from the system faster.
So if anyone gets irregular pulse; taking BHT immediately is a good idea.

http://www.vrp.com/b...350-mg-100-caps

I'll PM you the details now.

Thx Stefan

"...we show that in acutely damaged skeletal muscle, sequential interactions between multipotent mesenchymal progenitors and infiltrating inflammatory cells determine the outcome of the reparative process.

We found that infiltrating inflammatory macrophages, through their expression of tumor necrosis factor (TNF), directly induce apoptosis of fibro/adipogenic progenitors (FAPs). In states of chronic damage, however...macrophages express high levels of transforming growth factor β1 (TGF-β1), which prevents the apoptosis of FAPs and induces their differentiation into matrix-producing cells. Treatment with nilotinib, a kinase inhibitor with proposed anti-fibrotic activity, can block the effect of TGF-β1 and reduce muscle fibrosis....

Our findings reveal an unexpected anti-fibrotic role of TNF and suggest that disruption of the precisely timed progression from a TNF-rich to a TGF-β-rich environment favors fibrotic degeneration of the muscle during chronic injury."

I always believed that that Nilotinib would have system-wide effects due to its removing ailing Mitochondria and upregulation of autophagy in general.

That Nilotinib may be effective in the many Fibrosis related illnesses is very interesting!?

Nilotinib and liver fibrosis:

http://www.longecity...ndpost&p=767169

In summary:

Nilotinib was found to be "markedly" protective of liver fibrosis in livers challenged with highly toxic Carbon Tetrachloride at a HED of 284 mg/kg/day.

A restorative effect, at lower doses, may be expected when NOT trying to poison the liver!?

Logic,

Congrats on getting this project organized and executed.

I sent some info to an elderly friend who is still pretty sharp mentally but has a bad case of Parkinson's.

No response from either him or his son so maybe they will have an interest in a later group purchase.

Slightly off topic question though.  I've read your recommendation that BHT be available as a possible anti-dote to the Nilotinib if someone has a bad response.  Does that mean that someone who is regularly taking BHT should halt that product?  Or would taking the BHT with some time spacing before or after the Nilotinib be OK?

FYI I thought you would all like to know that logic has sent me 18 grams of Mebendazole for my dog with cancer.

It took a while as he had some care problems, but the Mebendazole has now cleared customs at the JHB intl Mail Centre and is about to fly out to the USA.

"Prevents amyloid secretion into the extracellular space between neurons." -- Actually, in Figure 3G, it appears to be reducing phosphotau by around 30% vs. DMSO control. This is much more useful for AD prevention than amyloid clearance. (Sufficiently early amyloid clearance might prevent AD, but in such situations, nilotinib would be difficult to justify as a prophylactic, given its side effect profile.) Impressive.

I have been walking a lot lately!

Progress Report:

I have just received an Email from the supplier saying that the payment reflects in their account (at last!) and that they will try to get the package sent (to the testing lab) on Thursday morning or Friday at the latest.

My apologies for the delay:
The PayPal fee would have been $ 229.18, so I decided to do a bank transfer.
The South African Reserve Bank gets very excited when more than R 50 000.00 leaves the country.
This, combined with the Easter break, that generally starts the weekend before Easter around here, didn't help.

The package will reach the testing lab next week.
That means your Nilotinib should arrive (USA) in around two weeks.

If you make another order for Nilotinib I might be interested. But what I'm worried about is how customs will deal with this. An envelope with a couple of grams in it will probably not be noticed, but a package with more than 10 grams might become a target for inspection and who knows what the consequences might be for bringing it in yourself. Aren't you guys worried about that?

SLEEPDEALER:

If concerned, a small amount such as 2,  5 or 10 grams an be sent in a few colored capsules or as powder to test the water. The postage will add up but the whole lot will not be confiscated. I don't dare do it here in Peru because I know from experience that they will steel anything just to do it, even if they throw it in the garbage later laughing. Ignorance is bliss AND creates some awful people. IMHO.

Hi Logic

Thanks for all your efforts.

If the test goes well, I will be up for the next lot.

I am prepared to travel to pick it up. I'm sure that customs would intercept it if it is in the post. As a sideline, Fedex may work, and that may be worth a try with a small amount to see what happens.

All strength to your bow!

David

Hi Sleepdaler
Glad to hear you are interested in future group buys of Nilotinib.

I did some research into getting this into the USA and worldwide and dont think it prudent to go into too much detail here, but:

Nilotinib is not a controlled substance:
http://www.drugs.com...-tki-10937.html

Also its being imported to a test lab in the USA where:

Products currently covered by valid US Patents can be ordered for R&D use in accordance with 35 USC 271 +A13(1)t

Hello David

Thx again for your kind donation which has been invaluable due to money transfer costs being much higher than anticipated.

I look forward to getting some Nilotinib from the next buy to you, one way or another!

Anybody interested in group buy #2 should post here, stating the amount they are interested in.

The package has cleared US customs and is on its way to the testing lab.   :)

I am currently sending amounts and addresses to the lab and organising with the USPS to supply the lab with Priority Mail Small Flat Rate Boxes for later pickup and final shipment. to all participating members.

Here is what your USPS package/box will look like:

Thanks for the tremendous effort despite so many impediments, Logic. As a result of all that, we have at least a chance that this may save lives! And yeah, tip of the hat to David Watford for his generosity.

If it fails the lab, that's life. We can always find another source. What matters is that you've demonstrated that this is at least theoretically possible to do. I think you should lead other group buys in the future, for which a reasonable management fee should be designated up front.

The only other request I have is that you remind the lab administrator of the importance of using Good Manufacturing Practices when dividing up the drug into individual packages. Among other things, this means (1) wearing disposable gloves and surgical masks and (2) avoiding contamination during the division process, for example due to debris on the scale or the knife or whatever. It's not so much about the bacterial hazard, as the possibility that we might end up giving people tiny amounts of very toxic substances which happen to have been tested at the same facility.

Thx RG  :)

I learned a lot in the process and any future buys will be faster without all the research I did and the practical mistakes and small oversights etc.
I would be happy to look at other group buys, besides Nilotinib, if those terms are ok with Longecity..?

Tiron looks very Interesting!?
http://www.longecity...idant/?p=763513

I don't think there will be an issue with the Nilotinib from my experience with the supplier and the fact that they know it's going to a lab for testing.

But if the purity is not up to scratch; the testing lab can further purify it by a process called recrystallization.

you end up with less product and it gets more pricy, but it's now in the country where most of this group buy's members reside...

GMP:

I discussed this important matter with the lab and they assured me that they will be following GMP:

Properly cleaned equipment, gloves, masks etc and it will be done by the same Dr. doing the testing.
