I previously ordered 50g of Nilotinib. I probably used ~0.4 grams (400 mg). I don't like the stuff and I don't think it's for me. I just don't think it agrees with my phenotype and my metabolism.

I'm pretty sure this drug works as advertised. I really noticed it working. But I just think the risks vs. the reward are not worth it for me.

If anyone wants my Nilotinib (50g) let me know via private msg. I am asking what I paid for it (~$500).

Hello all,

Here is my story: a Cortico-basal degeneration has been diagnosed on my mother a few weeks ago. The first symptoms appeared one year ago.

Now, the symptoms are changing very quickly to the point that, now, she can no longer walk.

The specialist tells us that there is currently no hope for this disease and that time is running out before total paralysis.

Does any of you have heard about a good effect of the N on this kind of disease?

Can I join a Group By even if I am In France? (where it is impossible to buy N).

My apologies for my absence; work has to come 1st:
I dashed back to Mozambique with a new gearbox and some other bits for the gold plant and some parcels for the people there.
Everything is running  smoothly... for now... and the rainy season normally puts a damper on operations,

Sorry to hear about your mother.

 

I did a very quick search and it would seem that the cause of Corticobasal Degeneration is Alpha-Synuclein and Tau etc.

This is the same as with PD and the other neurodegenerative diseases and means that Nilotinib may well be useful for treating it.

Here are the 3, yes 3, anonymous reports I could find in all the Emails and PMs sent to me.  (I feel there were more that I somehow missed while going through some 200 documents):

The reason for this is that Novartis holds a patent for Tasigna/Nilotinib. (Tasigna and Nilotinib are 2 names for the same thing, just in case there is any more confusion on this point)
This is why I had to 'jump through hoops of fire' to get the supplier to sell the stuff to me and the the reason for the disclaimer saying its for research & development purposes only.
ie: The supplier and the lab have serious and valid IP/legal concerns, as do many, and the only legal way to sell Nilotinib to people is for research and development.
This makes many reluctant to acknowledge owning and doing research with Nilotinib...!

  This video has an extremely interesting treatment for PD.  It uses 3 hormones and will be available in 3rd quarter of 2017.  It is a real eye opener and looks really good. It looks a lot better than Nilotnib.

Most likely for R&D into its senescent cell and old malfunctioning mitochondria killing abilities, as well as its anti fibrotic properties.  In short: Anti aging properties.
I have not, as yet, looked for, or come across any literature on the concomitant use of Nilotinib, Dasatinib and Quercetin , CLA etc.

While the effect is quite likely to be additive and quite possibly synergistic, the side effects may well be too..!?

If anyone has found any literature or has any experience in this regard; plz post.

However, since I've been taking 3,000 mg of EPA/DHA (from fish oil) a day I almost never have them.

 

However, from what you're saying, Logic, it seems prudent to avoid Nilotinib.

 

I've already paid for it, though.

My apologies for disappearing for a few days:
I ended up in Cape Town visiting family and bought a 3G dongle that refused to work wit my version of windows.  Its taken 3 days and another 3G dongle to get back online!

The order is placed and with the courier to the lab.

I did mention researching N..! :)

Perhaps a latecomer is interested in taking over your order?

Short of a doctor friend who can do the above checks and at whom you can take a dose; I wouldn't risk it.
The above effect may also only occur after daily use for a time, rather than a one off dose..???  Research reqd!  :)
I noted earlier in the thread that BHT upregulated CYP3A4 and may be a way to lower N levels, but wouldn't coun

Some suggestions from a lay-person
for preparing to take Nilotinib:

1. HEART

An EKG for arrhythmia is low tech, available at most little clinics.

Better:

From a Cardiologist, you can get more demanding tests:

1. "halter monitor" -- you wear it for 24 hr and on return, they analyze the recorded data for abnormalities.
2. Ultrasound/treadmill -- they run up your heart rate to see if you get arrhythmias under load and also video your valves in action for defects in functioning.

2. DRUG INTERACTIONS

•If you take any prescription drugs, go online and find the CYP 450 liver enzymes that metabolize them. Look at the list for 3A4 -- inhibitors and substrates all have to be discontinued--first. Notice that caffeine, cannabinol, grapefruit juice and theophylline (black tea and green tea) are also on the "no" list.

Don't take any N until you haven't taken anything metabolized by 3A4 for at least 2 weeks.

3. DOSAGE

•Its always a good idea to start low with any dosage since most side effects and adverse events are dose-dependent.

So if the target dose is 150 mg., one could start at 10 to 30 mg.; stay there for 2 weeks or more, and if uneventful, move up to 20 to 60 for another two weeks, and so on.

Even for a person beginning with 150mg., it is not realistic to expect improvements in 6 only weeks on a lifetime of accumulation (age 30-60=30 years!). Consider the dieter who thinks they can lose 20 lbs in a weekend. Biological processes can't be driven 100x faster in the opposite direction.

4. SUPERVISION

An Oncologist/Hematologist would be familiar with Nilotinib for Leukemia, but probably not neurodegeneration, so it's worth a try, but most people will be doing their own supervision.

See the Novartis website, other posts, for appropriate blood tests.

In addition, you need to keep a daily log of (at least) dosage, timing, activity level, mood, and symptoms to alert you to subtle patterns and changes that you could not possibly be aware of otherwise.

5. EMERGENCY

Keep your cellphone on you. Day and night. Always.

If at any time you feel odd, dizzy, a fluttering feeling in your chest, arm or chest pain--lie down on the ground wherever you are --breathe deep --and follow your pulse for consistent rhythm.

Call the paramedic rescue number (911 (US)) if there is the slightest doubt that you are perfect. Don't think twice about a false alarm. Nobody cares.

Has anyone heard of any interaction with rasagiline and N? I did notice that in the original Georgetown trial rasagiline was allowed, but the latest one (https://clinicaltria...how/NCT02954978) says " No mono-amine oxidase (MAO)-B inhibitors (Selegeline or rasagiline) are allowed at least 6 weeks before enrollment".
