GLYX-13 is a peptide.  Most peptides are produced in the form of salts - i.e. with a counterion.  TFA (trifluoroacetic acid) is the most typical counterion used for synthesis and HPLC purification of peptides.  If the counterion is not specified, it's likely TFA.  TFA is a corrosive acid and likely to be substantially more biologically toxic than acetic acid.  TFA should be removed / substituted with another counterion for anything that is going to be tested in vivo.

How would this fit with GLYX-13 analogues like NRX-1074? That's not a peptide.

I did not feel any effect from it, and I tried several different dosages. It could be very expensive taurine as the owner of Ceretropic concluded that several of their proprietary compounds were: https://www.reddit.c...esting_results/

I didn't feel any effects from ADD OX...so ceretropics findings are probably true.

Time to get this thread going again.

Seems that I had bacterial pneumonia without knowing it and when I'd insufflate the previous batch of glyx it would just make everything so much worse, thus the intense coughing, pain etc

OR Maybe just as likely: I snorted the previous (impure) batch of glyx and gave myself chemical pneumonia which then facilitated bacterial pneumonia. You have to remember that when injected, that batch of glyx caused major inflammation, burning, and major bruising at the site of injection and for several inches of the surrounding area.

Either way I'm done with snorting the glyx.

The take home message is that you should always keep the purity of any compound you're testing as pure as possible.

Morbidity and mortality are not compatible with longevity.

 

I found another supplier of glyx and tried a subcutaneous injection of the new batch dissolved in normal saline that was first passed through a 0.22 micron syringe filter.

There was no inflammation, burning pain, or bruising.

This new batch is what I'll begin reporting on.

And here is the report......

This batch of Glyx13 was as pure as can be found commercially.

Each vial was sterile and contained 200mg of powder.

I reconstituted the powder on one 200mg vials using sterile normal saline and then passed this solution through a 0.22 micron syringe filter just to be sure that it was sterile.

I injected a small amount of this solution subcutaneously in my upper leg with no pain or any issues. I waited maybe 1/2 hour to make sure there were no unanticipated reactions. There were no problems and so I injected the remainder of the 200mg vial. It did leave a very minor bruise at the injection site the next day.

Although I didn't expect and mood boost from this small dose, a mild boost did indeed occur and was especially evident the next day.

 

The second day after doing this, again with no issues, I began to wheeze when I inhaled and then began to feel pain under my ribs and in my abdomen.

Thees were the same exact side effects that I had experienced (and documented above) when I snorted the previous batch of Glyx that was far less pure.

It seems that I just react to the Glyx13 itself and all my previous theories about inhaling the stuff or lack of purity were wrong.

It's really too bad since this compound is extremely effective for my depression, which can get pretty intense, and unbelievably fast acting.

I am still experiencing some pain several days later and tried to preempt the major lung and pain side effects by taking ibuprofen, diphenhydramine, and 10mg of oral prednisone 3x per day (T.I.D.) as soon as I noticed the wheezing and pain starting up.

 

That's my report.

I think that Glyx13 has awesome potential, but for some reason I just have a recurring idiosyncratic reaction to the stuff.

Too bad.

Sorry to hear that. I hope this is not going to occure with everybody...

You say you get those symptoms after inhaling the medication; but you did

not get them after injecting it, did you ?

Perhaps this is just not supposed to be inhaled.

Have you tried to take it orally ?

I thought it was due to impurities in the first batch of "crude" Glyx13 as well as the method of administration...insufflating.

Turns out, in my case, it was simply a reaction to the Glyx itself.

Since Glyx is moving ahead in clinical trials, it must just me my personal physiological idiosyncrasies and wouldn't expect this side effect in many others based on Allergan's progress in bringing it through the approval process.

I paid just over $2k for 50 grams. I should have 48 grams left and you guys can have it at my cost if you want some.

I'd prefer to just unload the whole thing rather than parcel it out.

Sorry this is an error.

It should have been written:

I paid just over $2k for 50 vials of 200mg each, for a total of 10 grams. I should have 48 vials left.

 

Insufflation seems to be the most economical way to administer this compound based on my experiences, assuming no similar negative reactions to the glyx occur. This also assumes that the published dosages used in the clinical studies are accurate and are in the correct ballpark for future clinical use.

HPLC analysis shows greater than 98% for this synthesis with a tested value of 99.17%

I need to take a trip for a week, but I'll bring the Certificate of Analysis with me should anyone want to look at that.

Just a quick update:

Others have tried samples from this same lot I had trouble with, and have had no issues.

It seems that everything described above is my own personal side effect, though I would think that more people will also experience this as more try glyx.

Others should certainly be aware of this possibility, but that should not stop others from trying this compound if nothing else is working for them....in my opinion, and I am not a physician.

I have about 20 vials of GLYX-13 just sitting around in deep freeze, doing me no good.

They are 200mg and sterile. I paid $46 to have each vial synthesized.
