I have been taking RAPA for 2-3 months now, weekly about 6mg each time. I have not had any sore problems except for a silly mistake I made the first time. However, yesterday I decided to skip this week based on what Dare Devil mentioned to me in an email and I'm glad I did. From now on, I will skip a week every 5 weeks just in case there may be a slow buildup. Maybe that's the way we should all take it.

Took 5 mg. Sirolimus once a week for 3 months stopping 3 months ago. No negative effects, possibly increased inclination to exercise. Fasting glucose slightly higher, post-prandial excursion increased slightly, but A1c did not increase. Plan to repeat for 3 months starting next month.

Buying a drug from India certainly comes with some risk as well. Biocon appears to be a legitimate company and if dropshipmd really sends that brand maybe you are getting what you have ordered. But there have been problems with pharmaceutical companies from India (and other countries as well):

India appears to be a particularly risky place to source generic drugs.

My yearly physical results came back and LDL cholesterol now looks worrisome.

 

Besides rapamycin 2mg every 10 days I've started taking bergamot extract which is supposed to raise HDL while lowering HDL (and my HDL is quite a bit higher).

I've also been taking nicotinamide riboside 250mg and nicotinamide mono-nucleotide 250mg every other day, which is also supposed to lower cholesterol:

And also metformin 500mg/d and telmisartan 20mg/d

On my cardiologist's advice, I'm putting rapamycin on hold for a while to see if the LDL goes down, based on studies like this:

which indicated that rapamycin can increase cholesterol. That study also indicated that it increased triglycerides however, and mine have gone down. So we'll see what happens.

At this low dosage I don't believe Rapa is the culprit. Something else is at play.

DOB:  7-26-46  Male, 177 lbs., 71", Rapamycin 6 mg. once weekly for 16 months, (no side effects) Metformin 1000 mg. daily, N+R 1.5 gm of each + 15 gm Stearic Acid twice weekly, 5 mg Statin, Crestor daily. (will stop in 3 more months and get another lipid profile) BP 110-120/65-75. (measure daily)  Resting heart rate (52-56).  B/P and Cholesterol have dropped as I have lost weight.  No longer take B/P medication.  Have lost 20 lbs last 12 months.  Believe the combination of Rapa and Metformin helped.

 

Lab results 4-23-18: TC 130, LDL 74, HDL 43, Trigliceride 85,  Creatinine .5, GFR 108, Glucose 100, HbA1c 5.5 from 6.4 9 months ago. CBC within normal limits.

 

Had an arthritic rt. hip, no longer.  Can walk for many miles without any pain. Believe the rapa stops or slows down the arthritis deterioration.

I stopped taking it for a week and felt more awake (I thought) and took a dose today and feel sleepy. Could very well be a coincidence, but wondering if anyone else noticed something similar.

I stopped taking it for a week and felt more awake (I thought) and took a dose today and feel sleepy. Could very well be a coincidence, but wondering if anyone else noticed something similar.

 

In a word, yes.

 

Peak tiredness for me is normally on the 3rd day with weekly dosing, although recently this has changed and I'm experiencing this earlier. This might be because I've dropped back to a monthly dose; maybe this us something you get used to with weekly dosing, or maybe it is because the rest of my supplement regime has changed.

I was taking 5mg of rapamycin once a week, then cut back to every 10 days after 8 o r 9 months - total duration almost 11 months. I got a few mouth sores, my wife, who was also taking it, had frequent mouth sores and stopped around 3 months ago. 4 weeks ago I took a 5mg dose. The next day I was out in busy close company. I came down with some kind of flu the following day, and felt sick for 2 weeks. I have discontinued rapamycin for now. I have read that rapamycin may need not only to be taken intermittently, but for only a certain length of time. In the case of the middle aged mice, they were dosed for only 3 months I believe, and yet gained in health and l lifespan. Also,
if the half life of rapamycin is 60 hours, over an extended period there will be a slight build up of itinq the body, a very small and slow build up, but still some. I also want to add that we also take resveratrol, curcumin (not within a day or two of taking rapamycin) and NR, so it is not clear whether any of these other compounds also knock down mtor, and whether there is an net increase in effect.


NR will down regulate mTORC1.

Everolimus is the less toxic rapamycin analogue that binds the mTORC1 much more selectively.
